µpac columns – The microchip chromatography columns
Currently, many innovations across one industry are being employed to improve existing technology in another industry. One such example is the use of photolithography to generate microfluidics based nano lc columns by pharmafluidcs (Now under Thermo Fisher Scientific). Originally this technology was and is being used in the semiconductor industry. The advantage of this technology allows longer columns (up to 2 meters) to be generated in a 5 cm long chip. This configuration has benefits such as
1. Efficient quantitative analysis of low-abundant compounds
2. Higher resolution and therefore higher peak capacity during chromatographic separation resulting in the qualitative and quantitative analysis of 1000s of analytes present in a complex mixture. Pharmafludics columns are available in multiple configurations that are decided based on the i.d and length of the column. These configurations are run at different flow rates and chromatographic run times. The deciding factors for the selection of a particular configuration of columns are the complexity if a sample (no. of components) and the abundance of those components.
The µPAC columns are made by state of the art etching of a silicon wafer, and have a separation backbone of highly ordered and free-standing pillars. This unique design created exceptional properties resulting in excellent chromatographic performance, reproducibility and robustness. The µPAC columns are RP-C18 end capped, and compatible with any third party LC-MS system. They are ideally suited for application such as proteomics, metabolomics, lipidomics, biopharmaceutical peptide mapping and even single cell analysis.
